In silico doking, formulation and evaluation of quince and liquorice phytosomal nanosuspension for ulcers

Abstract
Peptic ulcer disease is a common gastrointestinal issue caused by an imbalance between harmful factors like gastric acid, pepsin, and Helicobacter pylori, and protective mechanisms
of the gastric lining. Traditional treatments can be effective but often lead to side effects and recurrence. This creates a need for safer and more effective options. This study looks at
developing a herbal phytosomal nanosuspension using quince seed (Cydonia oblonga) and liquorice (Glycyrrhiza glabra) to manage ulcers. The study included in silico molecular
docking of selected plant compounds against the ulcer-related target protein (PDB ID: 5YLU) using AutoDock Vina. Of the selected compounds, Amygdaloside had the highest binding
affinity at -8.0 kcal/mol, while Saringosterol had -7.7 kcal/mol, indicating strong anti-ulcer potential. Based on the docking results, the chosen herbal extracts were incorporated into a
phytosomal nanosuspension using the bottom-up nanoprecipitation method. The formulation was tested for sensory properties, pH, viscosity, particle size, and zeta potential. The
nanosuspension showed good physical stability, acceptable pH, uniform appearance, and satisfactory viscosity suitable for oral use. The combination of quince seed and liquorice
exhibited strong gastroprotective, anti-inflammatory, antioxidant, and healing properties. Therefore, the formulated phytosomal nanosuspension may offer a promising herbal method
for managing ulcers with better bioavailability and effectiveness.
